The estimation of kinetic constants for the lactate dehydrogenase system by the use of integrated rate equations.

The use of the integrated rate expression for the estimation of kinetic parameters has been tested with the lactate dehydrogenase-catalyzed reduction of pyruvate by reduced nicotinamide adenine dinucleotide at various pH values. The results show good internal precision and the results of duplicate determinations indicate that the results can be replicated satisfactorily. The values of kinetic constants estimated by this method show satisfactory agreement with those estimated from initial velocity measurements. Also, estimates of inhibition constants for oxamate and oxalate show reasonable agreement with estimates made from initial velocity measurements.
